Objective: To study the toxicity and toxicity comparison between Adriamycin liposome injection and Adriamycin injection after continuous intravenous injection.
Method: 90 SD rats were randomly divided into vehicle control group, adriamycin liposome injection (1 mg/kg) and adriamycin injection (1 mg/kg) group. Rats in each group were injected intravenously every 3 days for 13 consecutive times, and their body weight, blood biochemistry, hematology, organ indicators and other indicators were measured, and histopathological examinations were performed.
Result: Injecting 1mg/kg of Adriamycin liposomes can cause hair loss and ulcers in SD rats, significantly delay weight gain, increase PLT, ALT, BUN, decrease ALB, increase heart and kidney coefficients, and increase thymus. Testicular index decreased, histopathological examination showed that the testicular seminiferous tubules fell off, all levels of spermatogenic cells disappeared, some interstitial edema, cardiac myocardial fibers ruptured and dissolved and showed the disappearance of streaks and some nuclei, kidneys, histopathological lesions such as Tubular transparent tube is cast. Compared with doxorubicin liposome injection, doxorubicin injection can also cause the above-mentioned lesions at the same dose, but the rate of hair loss and skin ulcers in rats is reduced, and nephrotoxicity and cardiotoxicity are significantly increased.
Conclusion: Compared with conventional injection, Adriamycin liposome injection can significantly reduce the toxicity of the heart and kidney, but can significantly increase the toxicity of the skin.
