【Animal Experiment】-Based on OPG/RANK/RANKL system comparative study on bone metabolism characteristics of WHBE rabbits with forelimb deformities

  Purpose: Use OPG (osteoprotectin)/ANK (NF-κB receptor activator)/ANKL (NF-κBligand receptor activator) system to compare and study the bone metabolism characteristics of forelimb deformity WHBE rabbits.

  Method: Divide 10 WHBE rabbits, Japanese big-eared white rabbits and deformed WHBE rabbits into 3 groups, divided into HWR (healthy WHBE rabbits), HJR (healthy Japanese rabbits) and FMWR (predecessor WHBE rabbits) groups. .. Use the X-ray film taken on the X-ray machine to observe the radial shape of the forelimbs of each group of rabbits to determine the average gray value. The microscopic morphology of the forelimb bone tissue was analyzed by HE staining paraffin sections of bone tissue. Use fluorescence quantitative PCR method to detect OPG. The ANKL gene is expressed in the liver, and the expression of OPG/RANK/RANKL protein in serum and bone tissue was detected by enzyme immunoassay and immunohistochemistry.

  Results: Compared with the HWR and HJR groups, the ulnar radius of the forearm in the FMWR group was abnormally curved, the cortical bone was significantly thinner, and the gray value of the X-ray plain film was greater than that of the HWR group, and the gray value was extremely low (P\u003c0.05) . Rabbits in the FMWR group had liver RANKL gene expression level and RANKL/OPG mRNA ratio (P\u003c0.01), serum RANK, RANKL protein content and RANKL/OPG ratio (P\u003c0.05, P\u003c0.01), bone Tissue RANKL protein expression positive index, RANKL/OPG ratio (P\u003c0.05, P\u003c0.01) and other indicators were significantly higher than the HWR and HJR groups. Compared with the HJR group, the expression levels of OPG and RANKL genes in the liver of the HWR group were significantly increased (P\u003c0.05, P\u003c0.01).

  Conclusion: WHBE rabbits with deformed forelimbs reduced bone quality and damaged bone tissue. The ANKL/OPG ratio has increased significantly. Disorders of bone metabolism are the main cause of bone deformities. The difference in RANKL gene expression between white and Japanese big-eared white rabbits may be the induction of the susceptibility of WHBE rabbits to forelimb deformities.
