[Animal Experiment]-Synergistic effect of acupuncture on the treatment of osteoporosis in ovariectomized rats by transplantation of human amniotic membrane mesenchymal cells

  Objective: To explore the synergistic effect of acupuncture combined with human amniotic mesenchymal cell (HAMC) transplantation in the treatment of ovariectomized osteoporosis.

  Method: Select 45 non-pregnant Wistar rats and randomly divide them into sham operation group, model group and experimental group (acupuncture + human amniotic mesenchymal cell transplantation), 15 rats in each group. After the recovery of the surgical wound in each group, acupuncture and tail vein injection of HMC for 12 consecutive weeks, 7 consecutive days (except the sham operation group), the rats were sacrificed and sampled 24 hours after the last intervention. From the beginning of the intervention to the implementation, each group was weighed once a week. Dual-energy X-ray absorptiometry is used to measure the bone density at the midpoint, distal and proximal ends of the left femur, and the bone density is measured by atomic absorption spectroscopy. Calcium content; serum 25-hydroxy vitamin D (25-HVD), type I collagen C-terminal peptide (CTX-1), bone-specific alkaline phosphatase (BAP), and 12 weeks of intervention using ELISA in anti-tartrate The expression changes of enzyme 5b (TRAP5b) were detected in phosphate; after 12 weeks of intervention, the changes in biomechanical properties of each group of rats were detected. After 12 weeks of intervention, RT-PCR was used to detect the expression of transforming growth factor β1 (TGF-β1) gene in the spine.

  Result: The weight of the rats in the simulated operation group gradually increased with the natural growth of the animals. The model group started two months after modeling, but was much heavier than the sham operation group. The weight of the experimental group gradually increased, similar to the sham operation group. .. Compared with the model group, the bone mineral density and bone calcium content of the sham operation group were reduced, and the difference was statistically significant (P\u003c0.05). Compared with the model group, the bone mineral density and bone calcium content of the experimental group increased, and the difference was statistically significant (P\u003c0.05). The ELISA method showed that compared with the sham operation group, the expression of serum 25-HVD, CTX-1, BAP, and TRAP5b was significantly reduced, and the expression of the experimental group was significantly increased. (P\u003c0.05). In the biomechanical test, the ultimate load, ultimate stress and elastic modulus of the simulated operation group were significantly higher than those of the model group (P\u003c0.05), and the ultimate load and elastic modulus of the experimental group were higher than those of the sham operation group. It is also significantly higher (P \u003c0.05). T-PCR detection showed that compared with the sham operation group and the model group, the expression of TGF-β1m-RNA in the experimental group was significantly up-regulated (P\u003c0.05).

  Conclusion: Acupuncture combined with human amniotic mesenchymal cell (HAMC) transplantation has a synergistic effect on ovariectomized osteoporosis, and can improve the effect of osteoporosis in ovariectomized rats.
