Methods: The primary cultured 8d fetal rat hippocampal neurons were randomly divided into 6 groups: normal control group, H/R group, AI group, AI solvent control group, Bcl-2 inhibitor (TW-37) + H/R Group, TW-37 + AI + H/R group. Except for the normal control group, each group received 0.5 hours of hypoxia and hypoglycemia, and then 7 time points (0 hours, 0.5 hours, 2 hours, 6 hours, 24 hours, 72 hours and The index was detected within 120 hours. Cell viability was detected by MTT method, and the expression of caspase-3 protein and mRNA in hippocampal neurons was detected by Western blot and RT-PCR.
Result: Compared with the normal control group, the hippocampal neuron activity in the H/R group was significantly reduced at time 0 (P0.05).
Conclusion: Astragalus injection can enhance the activity of hippocampal neurons in H/R rats and reduce the expression of caspase-3 by activating the anti-apoptotic mechanism of Bcl-2. Bcl-2 is an astragalus injection that can inhibit hippocampal neurons in H/R rats.
